COMET-Test:<\/b><\/p>

Der COMET-Test ist eine Referenzmethode zur Bewertung der F\u00e4higkeit eines Inhaltsstoffs oder Kosmetikprodukts, DNA-Sch\u00e4den zu verursachen, einem wesentlichen Indikator f\u00fcr das genotoxische Risiko. In einer streng kontrollierten Umgebung werden im Labor gez\u00fcchtete Zellen dem Produkt ausgesetzt. Getestet unter Bedingungen, die eine realistische Exposition nachbilden.<\/span><\/span><\/span><\/span><\/p>\n

Der COMET-Test<\/span><\/span><\/span><\/span><\/strong> (oder <\/span><\/span><\/span><\/span>Einzelzell-Gelelektrophorese<\/span><\/span><\/span><\/span><\/em>) ist eine von der OECD (TG 489) anerkannte Referenzmethode zur Messung von DNA-Sch\u00e4den, die durch einen Inhaltsstoff oder ein Kosmetikprodukt verursacht werden. In einer Laborumgebung<\/span><\/span><\/span><\/span>Unter streng kontrollierten Bedingungen werden kultivierte menschliche oder tierische Zellen der zu testenden Substanz in realistischen Konzentrationen ausgesetzt. Verursacht die Verbindung DNA-Br\u00fcche oder -Ver\u00e4nderungen, fragmentieren diese Sch\u00e4den das genetische Material.<\/p>\n

Wenn die DNA besch\u00e4digt wird, fragmentiert sie. W\u00e4hrend der Elektrophorese wandern diese Fragmente aus dem Zellkern heraus und bilden eine charakteristische Struktur, die als \u00abKomet\u00bb bezeichnet wird: Der zentrale Teil stellt den Kopf dar (intakte DNA), w\u00e4hrend die besch\u00e4digten Fragmente den Schweif bilden. Die L\u00e4nge, Intensit\u00e4t und Ausdehnung dieses Schweifs werden genau gemessen, um das Ausma\u00df der Sch\u00e4digung der Zellen zu bewerten.<\/p>\n

Dieser \u00e4u\u00dferst empfindliche Test erm\u00f6glicht es, sehr geringe genetische Ver\u00e4nderungen nachzuweisen, die mit anderen Methoden manchmal nicht sichtbar sind. Er ist ein unverzichtbares Instrument zur \u00dcberpr\u00fcfung der toxikologischen Sicherheit von Rohstoffen, nat\u00fcrlichen Wirkstoffen und innovativen Formulierungen f\u00fcr den Schweizer und europ\u00e4ischen Markt, wo strenge gesetzliche Anforderungen an die Genotoxizit\u00e4t gelten.<\/p>\n

Technisches Prinzip<\/h3>\n

    Beschichtung der Zellen<\/strong> \u2013 Nach der Belichtung werden die Zellen in ein Agarose-Gel eingebettet.<\/p>\n<\/li>\n

    Lysat<\/strong> \u2013 Eine alkalische L\u00f6sung denaturiert die Membranen und setzt die DNA frei.<\/p>\n<\/li>\n

    Elektrophorese<\/strong> \u2013 Unter dem Einfluss eines elektrischen Feldes wandern die DNA-Fragmente aus dem Zellkern heraus.<\/p>\n<\/li>\n

    Entstehung des \u00abKometen\u00bb<\/strong> \u2013 Intakte DNA bildet die Kopf<\/em>, w\u00e4hrend die Fragmente wandern und eine Schwanz<\/em>.<\/p>\n<\/li>\n

    Analyse<\/strong> \u2013 L\u00e4nge, Intensit\u00e4t und Kopf-Schwanz-Verh\u00e4ltnis werden mithilfe von Bildgebungssoftware quantifiziert.
